Before being used for Intrauterine Insemination, semen must be processed to remove the seminal plasma. Seminal plasma contains prostaglandins which could cause severe contracts if placed in the uterus. The seminal plasma is removed by placing the semen on top of a discontinuous gradient and spinning in a centrifuge to bring the sperm to the bottom of the tube while leaving the seminal plasma at the top.
